A minimum of two peptide fragments are joined in CEPS. CEPS makes use of an enzyme featuring a cysteine in its active site giving it ligating properties. The initial fragment is synthesized with a C-terminal oxo- or thioester, which reacts with the active site cysteine of the enzyme to form a covalent thioester attachment, releasing organic alcohol. The addition of the second (amine) fragment replaces the thioester at the active site, linking to the first fragment to form the product. This leaves the enzyme fully functional again, to be reused for the next reaction. This cycle can continue with the product undergoing additional ligations
